测定意义:
                          
S-CAT是土壤微生物代谢的重要酶类,在H2O2清除系统中具有重要作用。

测定原理:

H2O2在240nm下有特征吸收峰,通过测定与土壤反应后溶液在此波长下吸光度的变化,即可反应S-CAT活性的高低。

需自备的仪器和用品:

紫外分光光度计、台式离心机、可调式移液器、1mL石英比色皿、研钵、冰和蒸馏水

Q;土壤过氧化氢酶:分光法的这个比色皿光径50px的行吗,还有这个湿土能做吗,还是必须风干?那风干37度要烘多久,不会对酶活性有影响吗?

A:使用比色皿光径50px的话计算公式中25px改为50px,湿土可以做,主要干土样品可以排除水分分布不均匀取样不均匀的问题,建议使用风干土样。一般37度隔1小时观察一下,风干较快,室温敞开风干过夜也可以。
